趋化因子受体CXCR4及其配体CXCL12在胃癌组织中的表达及其意义的论文 【摘要】 目的探讨cxcr4/cxcl12在胃癌组织中表达及其在肝转移中的作用。方法应用western blot 检测40例胃癌患者标本中肿瘤组织、邻近正常黏膜以及肝转移组织中cxcr4/cxcl12通路成员的表达情况，免疫组织化学法检测cxcr4/cxcl12在细胞水平的分布。结果胃癌组织中cxcr4/cxcl12表达水平明显增高（肿瘤组织vs正常黏膜,plt;0.05）；10例肝转移组织中cxcr4/cxcl12表达增高（转移组织vs原发肿瘤,plt;0.05）；cxcr4/cxcl12表达水平与tnm 分期晚（ⅲ/ⅳ）有关（plt;0.05）。结论cxcr4/cxcl12信号转导通路可能在胃癌肝转移过程中起一定作用，详细机制尚待进一步研究。 【关键词】 胃癌；信号转导；趋化因子；转移 expression and clinical significance of cxcr4/cxcl12 in gastric carcinoma ma xiangtao1,yu liwei1,fu jing2 department of surgery, department of pathology, beijing haidian hospital, beijing 100080,chinaabstract：objective to investigate the expression of cxcr4/cxcl12 and its potential role in hepatic metastasis of gastric carcinoma.methodsgastric cancer tissue, adjacent normal tissue and hepatic metastatic tissue samples were collected from 40 gastric carcinoma patients. several cxcr4/cxcl12 pathwayrelated proteins (cxcr4,cxcl12,cyclin d1 and bclxl)expression were detected by western blot. correlations between cxcr4/cxcl12 expression and clinicopathological parameters in gastric carcinoma were analyzed. cxcr4 and cxcl12 expression in cell level were determined using immunohistochemistry method.resultsthe expression of cxcr4 and cxcl12 protein significantly increased in gastric cancer compared to adjacent normal mucosa (plt;0.05), it was also highly expressed in metastatic tissue versus primary tumor tissue (plt;0.05) our data showed that expression of cxcr4 and cxcl12 protein are correlated with tnm colorectal cancer staging(plt;0.05).conclusioncxcr4/cxcl12 overexpressed in both primary and metastatic tissue of gastric carcinoma. it is implied from our results that cxcr4/cxcl12 signaling pathway plays an important role in hepatic metastasis of gastric cancer. key words：gastric neoplasm; signal transduction; chemokine; metastasis 1材料与方法 1.1材料 western blot中用的pvdf膜购自millipore公司，显影用的胶片购自柯达公司。www.11665.COm本研究中所有抗体购自santa cruz公司。预染标准分子量蛋白购自gibco/brl公司。ecl化学发光试剂盒来自amersham公司。浓缩蛋白分析液来自
