Marfan’s Syndrome 陈依然 Outline Introduction A systemic disorder of connective tissue caused by mutations in the ECM protein fibrillin-1. Autosomal dominant; locus to 15q21.1. The estimated prevalence is about one in 10,000. Cardinal manifestations include proximal aortic aneurysm, dislocation of the ocular lens, and long-bone overgrowth. *Clinical Manifestations Skeletal System Pectus carinatum（鸡胸） Pectus excavatum（漏斗胸） Reduced upper to lower segment ratio or arm span to height ratio 1.05 Positive wrist and thumb signs Ocular System *Ectopia lentis（晶状体异位） Abnormally flat cornea Increased axial（轴向长度） length of globe Hypoplastic iris（虹膜） or hypoplastic ciliary muscle（睫状肌） causing a decreased miosis（瞳孔缩小） Cardiovascular System *Dilatation of the ascending aorta at the level of the pulmonary sinuses *Dissection（剥离） of the ascending aorta Mitral valve prolapse Dilatation of main pulmonary artery Pulmonary System Skin Integument(覆盖物) Dural Ectasia（硬脑脊膜膨隆） Family/Genetic History *Pathology Fibrillin-1 is a main component of a class of 10–12 nm extracellular microfibrils found in a wide range of tissues such as the ciliary zonule. Microfibrils are thought to be necessary for both the formation of elastic tissues and to elastic properties of tissues themselves.
