PolyChromErythEM-FawcConc2-55.tif. * At left is an electron micrograph of an orthochromatophilic erythroblast (= normoblast), which is a maturing red blood cell that still has its nucleus. The nucleus is near the surface of the cell, in preparation for its extrusion. The electron micrograph at right shows the process of extrusion. After the nucleus is released, the cell is an erythrocyte. The macrophage of the blood island phagocytoses the extruded nucleus. From the Erlandsen slide set (MH). MH-5E1. Reticulocytes are slightly-immature red blood cells, in which the last remnants of cytoplasmic ribosomes, mitochondria and other organelles still persist in what is otherwise a concentrated solution of hemoglobin. Reticulocytes are released into the blood, and become mature erythrocytes in about 24 hours. A count of reticulocytes in a blood smear is a measure of new RBC production. It is often difficult to distinguish reticulocytes from mature erythrocytes in ordinary blood smears. However, the blood that was to be used for this smear was treated with a basic dye, brilliant cresyl blue, which stains the RNA blue in clumps of ribosomes, and then the smear was made. The staining pattern in the cells looks a little bit like a net, giving the cell its name (Latin, reticulum=fishing net; Greek, cyte=cell). From Wheaters Functional Histology, 4th edition, page 48.
