PAGE PAGE IV Transcriptional profile of primary chicken hepatocytes responsed to growth hormone Abstract Transcriptional profile of primary chicken hepatocytes responsed to growth hormone Abstract Growth hormone (GH) can affect the growth of animal body by regulating the metabolism of three major nutrients. Since the first published data on the metabolic effects of growth hormone 50 years ago, more and more researchers have focused on it. And the study of genes which are regulated by growth hormone seems more important than others. In recent years, with the development of next generation sequencing technology, scientists use it almost in all aspects of biology, of which, transcriptome sequencing caused peoples attention for it plays an important role in the study of gene expression. In this study, we isolated primary hepatocytes from male AA chickens (4~5 weeks old) using modified two-step semi-situ collagenase perfusion method combined with the homogenate digestion process. After cultured with Williams E medium, the cells had a good morphology. After isolating hepatpcytes successfully, we treated the cells with chicken growth hormone (cGH), and the result revealed the best time to treat the hepatpcytes is 12 to 24 h. At the same time we researched the relative expression of some gene family members, we found that the expression level of SOCS family except SOCS-1 was up regulated after growth hormone treatment. However, the expression level of HNF gene family’s members including HNF-1α、HNF-1β、HNF-3β did not changed after growth hormone treatment. We used transcriptome sequencing to further study the genes regulated by growth hormone. The result indicate that there were 164 genes differently expressed after growth hormone treatment, 112 of them were up regulated and 52 were down regulated.
