北京工商大学毕业论文(设计) PAGE  PAGE III 编号:1002070321 毕业论文(设计) 题目气相色谱法测定大鼠粪便短链脂肪酸色谱条件的优化 院(系) :食品学院 专业 :食品科学与工程 学生姓名 : 成绩: 指导教师肖俊松(职称)副教授 2014年05月 PAGE \* MERGEFORMATII 摘 要 气相色谱法是测定短链脂肪酸的常用方法。为了研究原花青素对肥胖大鼠肠道菌群的影响,需要对大鼠粪便中的短链脂肪酸定性和定量。本论文对影响短链脂肪酸测定的两个关键因素:样品前处理方法以及色谱条件进行了优化,比较了三种色谱条件以及四种样品前处理方法:盐酸水提法、偏磷酸处理法、水提乙醚萃取法和固相微萃取法。结果表明最优样品前处理法为盐酸水提乙醚萃取法,优化的色谱条件为:色谱条件B。色谱条件B:柱型号JM122-7032;柱规格为30.0m×250mm×0.25μm的DB-WAX毛细管柱。测定采用程序升温,初温100℃,保持0.5min;以8℃/min速度升至180℃,保持1min;然后以20℃/min速度升至200℃,保持5min,分流比为10:1,载气为He,流速为1.2mL/min,FID检测器温度为250℃,进样口温度为250℃。测定了15个大鼠粪便样品中短链脂肪酸的含量。 关键词:短链脂肪酸;大鼠粪便;气相色谱;样品前处理 ABSTRACT Gas chromatography is a common method for the determination of short-chain fatty acids.To study the effect of proanthocyanidins on gut microbiotaof rats,the short-chain fatty acids were qualified and quantified.Two key factors influencing the determination of short-chain fatty acids,i.e.,sample preparation methods and chromatographic conditions,were optimized,and totally3 chromatographic conditions and 4 sample preparation methods were compared,i.e.,hydrochloric acid-acidified water extraction,metaphosphoric acidextraction, acidified water extraction followed by ether extraction and solid phase micro-extraction.Results showed that optimum preparation method was hydrochloric acid-acidified water extraction,and the optimum GC conditions were as follows:Column Type JM122-7032;30.0m×250mm×0.25μm the DB-WAX capillary column.Determined by temperature-programmed,initial temperature 100℃,keeping 0.5min;with 8℃/min rate rose to 180℃,keeping 1min;then with 20℃/min rate rose to 200℃,keeping 5min,split ratio of 10:1, The carrier gas is He,flow rate 1.2mL/min,FID detector temperature is 250℃,inlet temperature is 250℃.Totally 15 rats feces samples were determined.
