AbstractPotatolateblightisadevastatingdiseasewhichleadstotherotofpotatoleaf,stemandtuber.Currently,cultivationresistantvarietyderivedfromplantresistantgeneisthefirsttacticforcontrolthedisaese.Inthisstudy,weusedthreelateblightresistancegenes,whichbelongtothesamegenefamilyofCC-NBS-LRRtype,sharedwithabout82%identitieswithaminoacidsequence.BothRpi-mcq1.1andRpi-mcq1.2wereclonedfromSolanummochiquense,andRpi-vnt1.1wasclonedfromSolanumventurii.BychangedwithRpi-vnt1.1genepromoter,domainswappingoftheRpi-mcq1.1andRpi-mcq1.2,wegoaltoseekafterresistanceimprovementandtoexplorethedomainthatcontrolledtherecognizationbetweenRgeneandpathogeneffector.Firstly,threenewconstrutshavebeenmadewithoverlappingPCRtechniqueanddomainswaptactic.TheV1M2wascombinatedwithCC-NBmotifofRpi-vnt1.1andLRRmotifofRpi-mcq1.1.RpiV::mcq1.1wasmadebyreplacedthenativepromoterwithRpi-vnt1.1promoter.RpiV::mcq1.2wasmadebyreplacedwithRpi-vnt1.1promoterasRpiV::mcq1.1.ThreenewgeneswereligatedintopCAMBIA1300,andmobilisedintoAgrobacteriumstrainAglⅠ,thenintroducedintosuceptiblepotatovarietyDesireeandNicotianabenthamiana.ForeachconstructV1M2,RpiV::mcq1.1andRpiV::mcq1.2,13,11and25positiveindividualswereconductedwithpotatoDesireeregardedas73%,61%and79%oftransformationefficiencyrespectively.Theamoutof8,7and13positiveindividualswereconductedinN.benthamianawith37%,30%and41%oftransformationefficiencyrespectively.AllthematerialswillbetestedforinoculationPhytophthorainfestans,totestwhethertheirresistantspectrumhavechangedornot,andtodetectwhethertheyhavebeenaffectedtorecognizetheAvr2.ByusingAgrobacteriummediatedtransientexpressionsystemonN.benthamiana,Rpi-mcq1.1,V1M2,RpiV::mcq1.1andRpiV::mcq1.2wereindicatedtorecognizetheAvr2(G3,G12)afterco-infiltrationwithGV3101carriedeachresistancegeneandAvr2.TheresultssupporttomakeaconclusionthatitisLRRmotifofRpi-mcq1.1correspondfortherecognizationbutnottheCC-NBSmotif.Moreinterestingly,wefoundthatRpi-mcq1.2wasturnstorecognizetoAvr2afterreplacedwithRpi-vnt1.1promoterwhichcouldn’ttriggerHRonN.benth
