TheresearchoftheinfluenceofAjugadecumbensthunbonthegrowthofhemangiomaandtheexpressionofFas/FasLinnudemicemodelAbstract:Objective:InordertoobservetheinfluenceofAjugadecumbensthunbonthegrowthofhemangiomaandtheexpressofFas/FasLinnudemicemodel,anddiscusstheeffectandthemechanismofAjugadecumbensthunbasatreatmentforhemangioma.Methods:Infantproliferativehemangiomawastakenbysurgeryandseparatedintosmallpieces(3mm*4mm*5mm)understerileconditioninoperatingdepartment.Then,tissuepieceswereimplantedsubcutaneouslyintospecificpathogenfreenudemice,eachmousewasimplantedtwotumortissuepieces.30dayafterimplantation,wehad65nudemicethathemangiomasurvivedandgrewnormally.Randomlyselected5tobemeasuredthevolumeofhemangiomaandthensacrificed,thehemangiomatissuesweretakenoutandmadeintoparaffinsections,asprimarycontrast；remaining60nudemicewererandomseparatedintothreeexperimentalgroupstodorandomizedcontrolledtrails：vaselinegroup,Ajugadecumbensthunbgroupandtriamcinoloneacetonidegroup,thatweremeasuredthevolumeofhemangiomaandgivendifferentmedicine.Eachgrouphad20nudemiceand40tumors.Linimentofvaselineanddimethylsulfoxidemixtureascontrolchemistrywasadministratedeverydayinvaselinegroup.Linimentofself-madeAjugadecumbensthunbwasadministratedinthesurfaceofhemangiomaeverydayinAjugadecumbensthunbgroup.Triamcinoloneacetonidewasinjectedintothehemangiomaonceatthebeginningintriamcinoloneacetonidegroup;thedoseoftriamcinoloneacetonidewas0.05ml(4mg/ml).Themiceinthreegroupswerefedequally.Afterbeadministrateddifferentmedicine,thegrowthandstatusoftumorwasobserved.Inthemeantime,thevolumeofhemangiomainthreegroupswasmeasuredinvivo.After7day、14day、21dayand28day，randomlyselectedfiveexperimentalmiceineachgrouptobesacrificed,thehemangiomatissuesweretakenouttobemeasuredthevolumeandmadeintoparaffinsections.Hematoxylin-eosin(HE)stainingofallparaffinsectionsandanalyzedthehistologicalchange.ImmunohistochemistrystainingofallparaffinsectionswithFas、FasLandcysteinylaspartatespecificproteinase8(caspase-8)antibodyandanalyzedtheirexpress.
