PreparativeSeparationofStilbeneGlycosidesandFlavonesFromtheBranchofMulberryAbstractMulberry(MorusalbaL.)isaneconomicagroforestrytreewhichhasbeenexploitedinthesericultureindustry.Differentpartsofthisherb,suchastheramulus,leavesandcortexofMulberryarealsoappliedastraditionalChinesemedicinalherbspossessingmultiplemedicinalfunctions.MulberrosideAandotherstilbeneglycosidesexistedincortexofMulberry,whichareknownasstrongantioxidantsbeneficialforhealth.MulberrinandotherflavonoidswerecharacteristicconstituentsintheramulusofMulberrywhichareresponsibleforitsanti-inflammatoryactivities.Oldbranchofmulberryshouldbeprunedandgenerallydiscardedforbettergrowthofleaves.Recently,itisreportedthatsomestilbeneglycosidesandflavonoidsexistedinbranchofmulberry.Inthisstudy,weaimtoclarifythepresenceoftheseconstituentsinthebranchofmulberryusingHPLCanalysis.Apreparativeseparationprocessofbothstilbeneglycosidesandflavonoidsfractionswouldbedevelopedforthefirsttimeusingmacroporousresin,whichmaybehelpfulforfurtherexploitationofthismaterial.Aseparationprocessofbothstilbeneglycosidesandflavonoidsfractionsfromthebranchofmulberrywasdevelopedusingmacroporousresin,usingMulberrosideAandMulberrinaschemicalindicators.Bothstatic/dynamicadsorptiontestswereundertakentoevaluatethemaximumadsorbingcapacities,desorptionratiosandseparatingresultsofresins.AB-8resinwasselectedasoptimaloneamong9typicalresinsinvestigated.FurtherstaticadsorptiontestsrevealedthatAB-8fittedwiththepseudo-second-orderkineticsmodelandLangmuirisothermmodel,respectively.Dynamicabsorption/desorptionexperimentsonAB-8columnswereconductedtoobtaintheoptimalseparationparametersasfollows:thefeedvolumeoftheextractsofthebranchofH2mulberry(MA-1,2.0gherbalmaterial/mL)wassettobe1bedvolume(BV)ofAB-8column,theadsorptionequilibriumtimewas60min;elutionfirstwithwater(3BV),followedby40%ethanol(4BV,MA-2),finallyby90%ethanol(7BV,MA-3)insuccess;andstilbeneglycosidesandflavonoidsfractionswereobtainedfromMA-2andMA-3eluentes,respectively.Throughonlyonecyclescale-uptrea
