PAGE PAGE IV The effects of Icariin on MC3T3-E1 cell cultured with titanium particles in vitro Abstract The effects of Icariin on MC3T3-E1 cell cultured with titanium particles in vitro Abstract Objective Icariin has been reported to stimulate proliferation, differentiation and osteogenic function of osteoblasts, yet little is known whether Icariin can inhibit the effect of titanium(Ti) particles on osteoblasts. The aim of this study was to investigate the effects of Icariin on the pre-osteoblastic MC3T3-E1 cell cultured with titanium particles by studying the proliferation, differentiation and gene expression . Methods MC3T3-E1 cells were incubated with diferent concentrations of ICA(10-7-10-5 M) and titanium(Ti) particles. CCK-8 method was used to observe the cytotoxicity of ICA and the effect of Icariine on cell proliferation. The activity of alkaline phosphatase (ALP), TypeⅠcollagen(COLⅠ) and osteocalcin(OCN) were assayed to observe the effect on cell differentiation by Enzyme-linked immunosorbent assay(ELISA). The amount of calcified nodules were assayed to observe the effect on cell mineralization. The expression of Osterix(OSX) and Runt-related transcription factor 2 (Runx-2) mRNA was detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Results The cytotoxicity assay showed that icariin(10-7-10-5 M) had noncytotoxicity. To study the effects of Icariin on cell proliferation, MC3T3-E1 cell were cultured in the medium with Ti particles(0.1mg/ml) and different concentrations of icariin for 1, 3 and 7 days. Icariin(10-7-10-5M) promoted (P0.01) the Ti-inhibited cell proliferation compared to the controlled group on the 3rd and 7th day after being cultured with Ti(0.1 mg/ml), but the significant inhibitory effect (P0.01) on cell proliferation was achieved on the 3rd and 7th day when cells was exposed to Icariin(10-4 M) and Ti(0.1 mg/ml).
