健胃愈疡颗粒对大鼠胃粘膜癌变DNA含量TNF-α基因表达.DOCVIP

红景天对大鼠胃粘膜C-myc表达及细胞凋亡的影响 黄国栋1△, 黄媛华2，黄道富2, 肖美珍2,唐丽君 3 （1南昌大学第一附属医院，江西省南昌市 330006；2湖南中医药大学附属衡阳医院，湖南省衡阳市 421001;3湖南省人民医院，湖南省长沙市 410000） 【摘要】　目的　研究红景天加维甲酸对大鼠胃粘膜癌变DNA 含量、C-MYC mRNA 表达及细胞凋亡的影响。方法　采用MNNG、酒精及10 %NaCl 混合液建立大鼠胃癌模型,设立正常空白组、模型组、 维甲酸治疗组和红景天加维甲酸治疗组,分别采用Feulgen 染色、免疫组化,Western blotting 及TUNEL 法检测大鼠胃粘膜DNA 含量、C-myc基因表达及细胞凋亡。结果　红景天加维甲酸治疗组癌变发生率低于模型组和维甲酸治疗组。C-myc基因表达与细胞凋亡受抑制和胃粘膜癌变的发生,呈递增趋势,并且两者具有一定的相关性。结论　红景天加维甲酸抗癌,下调C-myc基因表达,促进细胞凋亡是其重要机制之一。 【关键词】红景天;胃肿瘤; 动物; C-myc; 细胞凋亡 Effects of Gold Theragran on C-myc gene exepression and apoptosis of gastric cancer in rats Guo-Dong Huang1， Yuan-Hua Huang2 ，Dao-Fu Huang2，Mei-Zhen Xiao2，Li-Jun Tang3 （1the First Affiliated Hospital of NanChang University , NanChang, JiangXi Province 330006; 2 the Affiliated Henyang Hospital,Hunnan University of Traditional Chinese Medicine,Henyang City ,Hunan Province,421001; 3the Peapole’s Hospital of Hunan Province, Changsha City , Hunan Province，4l0002） 【Abstract】　Objective 　To study the effects of Gold Theragran combined retinoic acid on DNA ,C-myc exprssion and apoptosis of gastric cancer in rats. With the development of gastric cancer , the expression of C-myc gene and supression of apoptosis were increased. A corrolation was present between both. Conclusion Gold Theragran combined with retinoic acid can both downregulated the expression of C-myc gene and promote
