PAGE 攀枝花学院本科毕业设计（论文） 金龙胆草中总皂苷的提取研究 学生姓名： 学生学号： 院（系）： 年级专业： 指导教师： 二〇一二年五月 攀枝花学院本科毕业设计（论文） 摘要 PAGE 20 摘要 本实验采用水浴回流提取，分别通过单因素实验、正交实验考察金龙胆草总皂苷的最佳工艺条件，为金龙胆草总皂苷药理药效研究提供材料，同时为其进一步开放奠定基础。方法 以熊果酸为对照品，用紫外可见分光光度法建立熊果酸的标准曲线；在单因素试验的基础上，以总皂苷含量为指标，采用正交试验方法考察提取温度、固液比、乙醇浓度、提取时间等因素对提取工艺参数做初步研究。结果　（1）单因素实验结果：设计金龙胆草的提取工艺为乙醇浓度分别为50%，60%，70%，80%，90%；固液比分别为1：20，1：25，1:30，1：35，1：40；提取温度分别为50℃，60℃，70℃，80℃，90℃；提取时间分别为0.5h，1h，1.5h，2h，2.5h；（2）正交实验结果：提取温度A，固液比B，乙醇浓度C对指标影响显著。根据以上分析，可定出影响指标的因素显著性主次顺序为：ABCD。总皂苷的优选浸提条件（各因子优水平组合）为最佳提取温度70℃，，最佳固液比1:30，最佳乙醇浓度为80%，最佳提取时间2h。 关键词：金龙胆草； 总皂苷；水浴回流提取； 正交试验法 攀枝花学院本科毕业设计（论文） ABSTRACT ABSTRACT The purpose of Water bath reflux method extraction process of total Saponins in Gold gentian conditions of a preliminary study and with the traditional craft make that nol compared to optimize the extraction of total Saponins in Gold gentian conditions.
