※生物工程 食品科学 2007, Vol. 28, No. 04 231 1 2 2 2 1 1 路玲玲 ,王 敏 ,朱会霞 ,孙金旭 ,檀建新 ,张 伟 (1.河北农业大学食品科技学院生物工程系,河北 保定 071001; 2.衡水学院生命科学系,河北 衡水 053000) :选择化学药物和紫外灭活单亲原生质体为遗传标记,以0.01%的三唑酮和紫外照射h-1原生质体2min为 遗传标记,采用四因素三水平正交试验确定了融合子LZ-3的浓醪酒精发酵工艺。浓醪发酵的最佳工艺条件是:料 水比为1:2,糖化酶用量为 400U/g 原料,接种量为 9%(V/V),初始pH值为5.5,在此条件下,以玉米粉的糖化醪 为培养基,发酵72h,LZ-3 的酒精产率为9.0%。 :原生质体;融合;耐高温;高产酒精酵母 Building Thermotolerant High-alcohol-producing Yeast by Proplast Fusion 1 2 2 2 1 1 LU Ling-ling ,WANG Min ,ZHU Hui-xia ,SUN Jin-xu ,TAN Jian-xin ,ZHANG Wei (1.Department of Biology Engineering, College of Food Science and Technology, Agricultural University of HeBei, Baoding 071001, China;2.Department of Life Science, Hengshui University, Hengshui 053000, China) Abstract :As chemical drugs and UV used to exterminate the single parent protoplasts were choosen as hereditary markers, with 0.01% triadimefon content and irradiated by UV 2 min, the optimum conditions of the orthogonal test are: 33.3%(W/V) concentration of crude corn in fermenting material, 400 U/g of glucoamylase (per gram of raw corn), 9%(V/V) inoculum of yeast, and initial pH 5.5. The orthogonal test indicated that the fusion strain LZ-3 produces the highest amount of alcohol, or 9.0% alcohol after incubation at 42℃ for 72 hours.
