六味地黄汤辅助环磷酰胺对荷H22肝癌小鼠MMP9、NF-KB表达的影响 饶斌,谢斌,谢雄,余功,刘红宁资助 资助项目：国家自然科学基金资助项目 ，江西省研究生创新基金项目（YC2014-S275) ，江西中医药大学研究生创新基金项目（JZYC14A02) 第一作者：饶斌，硕士研究生，从事中医药抗肿瘤研究。E-mail: 837585272@ *通讯作者：刘红宁，博士生导师，教授。主要研究方向：中医基础理论研究。E-mail:lhongnin@ (江西中医药大学，江西 南昌 330004) 摘要 目的：观察六味地黄汤辅助环磷酰胺对荷H22肝癌小鼠MMP9、NF-KB表达的影响。方法：雄性昆明小鼠40只，随机分为模型组、化疗组、六味组、六味化疗组，每组10只。腋下注射H22细胞建立H22荷瘤模型，六味组造模前两周开始用药（剂量为22g·kg-1·d-1），化疗组（剂量为50mg·kg-1·d-1,隔日1次）及六味化疗组造模后用药，造模14d后处死小鼠称重，取瘤组织称重计算抑瘤率，取脾组织称重计算脾指数，免疫组化检测MMP9、NF-kB蛋白的表达。结果：化疗组及六味化疗联合组具有明显的抑瘤作用，与模型组比较有统计学意义（P0.01），与六味组比较，有统计学意义（P0.05）；六味组脾指数增大，与模型组及化疗组比较，均有统计学意义（P0.05），六味化疗联合组的脾指数均高于化疗组，有显著性差异（P0.01）；与模型组比较六味化疗联合组MMP9蛋白表达水平显著下降（P0.05），六味、化疗、六味化疗联合均可显著降低NF-kB蛋白的表达水平，与模型组比较，有统计学意义（P0.05或P0.01）。结论：六味地黄汤辅助环磷酰胺可提高动物免疫功能，其抗肝癌的协同机制可能不是通过NF-KB，而是通过降低肝癌组织MMP9蛋白表达来实现。 关键词：六味地黄汤；环磷酰胺；肝癌；MMP9；NF-KB Effect of Liuwei Dihuang decoction?adjuvant cyclophosphamide on MMP9,?NF-kB expression in H22 tumor-bearing mice RAO Bin, XIE Bin, XIE Xiong, YU Gong, LIU Hong-ning* (Jiangxi University of Traditional Chinese Medicine,Jiangxi Nanchang 330004 China) Abstract : Objective To observe the?effect of Liuwei Dihuang decoction(LW) adjuvant cyclophosphamide(CTX) on MMP9,?NF-kB expression in H22 tumor-bearing mice. Method 40 male Kunming mice were divided into model group, CTX group, LW group and the group using the LW and CTX,each group contained 10 mice. Animal models of H22 tumor- bearing mice was induced with the axillary?injection of H22 cells, LW group started?medication at molding?two weeks before(22g/kg?d), CTX group(50mg/kg?2d) and the combination group started?medication after molding. After 14d modeling, kill the mice and weigh, weighing the tumor tissue and splenic tissue, calculation of the antitumor effect and spleen index, testing the expression of MMP9,NF-kB protein on immunohistochemistry assay.
