凋膜止崩液对体外培养人子宫内膜.PDFVIP

国际中医中药杂志2018年11月第40卷第11期 Int J Trad Chin Med, November 2018, Vol. 40, No.11                           ·1065 ·  ·实验与方药 · 凋膜止崩液对体外培养人子宫内膜 腺上皮细胞 Bcl-2/Bax 蛋白及基因 表达的影响 李楠 徐翠 贺丰杰 谈勇 王灿灿 【摘要】 目的 观察凋膜止崩液对体外培养人子宫内膜增生症患者内膜腺上皮细胞 Bcl-2/Bax 蛋白 及 mRNA 表达的影响。方法 收集病理诊断为子宫内膜增生症患者的子宫内膜组织，经分离、培养、鉴 定后，按照随机数字表法将腺上皮细胞分为分为空白对照组，凋膜止崩液低、中、高剂量组。空白对照 组加入含 10%FBS 培养液，凋膜止崩液低、中、高剂量组在含 10%FBS 的细胞培养液中分别加入 12.5、 25、50 μg/ml 的凋膜止崩液。干预后48 h，采用蛋白免疫印迹法及RT-PCR 法检测腺上皮细胞 Bcl-2 、Bax 蛋白及 mRNA 表达。结果 与空白对照组比较，凋膜止崩液低、中、高剂量组 Bcl-2 mRNA [(1.51±0.07)、 (1.09±0.06)、(0.93 ±0.07)比(1.92±0.08)]及蛋白[(0.91 ±0.02)、(0.72 ±0.03)、(0.62 ±0.03)比(1.28± 0.02)]表达降低(P ＜0.05)，Bax mRNA [(5.73 ±0.37)、(8.00 ±0.23)、(10.72±0.40)比(3.27 ±0.34)]及蛋 白[(0.45 ±0.02)、(0.63 ±0.02)、(0.78 ±0.03)比(0.32 ±0.02)]表达升高(P ＜0.05)。与低剂量组比较，中、高 剂量组 Bcl-2 mRNA 及蛋白表达明显降低(P ＜0.05)，Bax mRNA 及蛋白表达明显升高(P ＜0.05)。结论 凋 膜止崩液可增加腺上皮细胞对凋亡的易感性，促进细胞凋亡。 【关键词】 子宫内膜增生；细胞凋亡；基因，bcl-2 ；bcl-2-相关 X 蛋白；凋膜止崩液 Effects of the Diaomo-Zhibeng liquid on the expression of Bcl-2/Bax mRNA and their protein in vitro human endometrial glandular epithelial cells * Li Nan , Xu Cui, He Fengjie, Tan Yong, * Wang Cancan.
