英文摘要 妊娠期高盐饮食重编程血管紧张素系统介导成年子代心肌凋亡 PART2 The pro-apoptosis effects of maternal high-salt diet during pregnancy on myocardium and the intracellular signaling pathways [Objective] To investigate the relationship between RAS and apoptosis, in vivo and in vitro, as well as possible cell signal pathways. [Methods] TUNEL was used to determine the apoptosis body in the myocardial tissue of adult offspring quantitatively. Apoptosis in the heart tissue was detected with DNA ladder. H92C cells were pretreated with different concentrations of Ang Ⅱ, CGP42112A(AT2 agonist), then Losartan(AT1 antagonist), and PD123319(AT2 antagonist). Mitochondria and cytoplasm were separated and measured with western-blot to determine the possibly involved cell signal pathways. [Results] The expression of apoptosis body was higher in the myocardial tissue of adult offspring in HS group. In vitro, H9C2 cell line, cardiac myoblasts were introduced more apoptosis by adding Ang Ⅱ and CGP42112A(AT2 agonist). This apoptosis could be reversed by PD123319(AT2 antagonist), not losartan(AT1 antagonist) . Caspase-3 and cytochrome C were higher in the cytoplasm and lower in mitochondria. [Conclusion] Maternal high-salt diet during pregnancy may elevate the expression of AT2 in the heart of offspring,and induce myocardial apoptosis.
