中草药 Chinese Traditional and Herbal Drugs 第47 卷 第8 期 2016 年4 月 ·1361 · 蜘蛛香提取物及总缬草素的抗焦虑活性研究 # * * 翟 欣，孔周扬 ，王素娟，蒋益萍，张 宏，郑承剑 ，秦路平 第二军医大学药学院 生药教研室，上海 200433 摘 要：目的 探讨蜘蛛香提取物及蜘蛛香总缬草素的抗焦虑作用及相关机制。方法 利用小鼠明暗箱、敞箱、高架十字迷 宫和自发性活动实验观察小鼠行为学方面的表现，评价蜘蛛香提取物及总缬草素抗焦虑作用；利用 ELISA 试剂盒方法测定 小鼠血清皮质酮和脑海马组织中神经递质5-羟色胺 （5-HT ）、去甲肾上腺素 （NE ）和多巴胺 （DA ）水平。结果 蜘蛛香提 取物及总缬草素各剂量组对小鼠在明暗箱、敞箱、高架十字迷宫和自发性活动实验中的行为学表现均有改善作用，且可降低 小鼠血清皮质酮水平和下调脑海马组织中神经递质NE 、DA 及5-HT 的水平。结论 蜘蛛香提取物及总缬草素均有抗焦虑作 用，其抗焦虑作用可能是通过作用于丘脑-垂体- 肾上腺 （HPA ）轴系统和调节脑组织神经递质而实现的。 关键词：蜘蛛香；总缬草素；抗焦虑；皮质酮；神经递质 中图分类号：R285.5 文献标志码：A 文章编号：0253 - 2670(2016)08 - 1361 - 05 DOI: 10.7501/j.issn.0253-2670.2016.08.019 Study on anti-anxiety activity of extract and total valepotriate in Valerianae Rhizoma ZHAI Xin, KONG Zhou-yang, WANG Su-juan, JIANG Yi -ping, ZHANG Hong, ZHENG Cheng-jian, QIN Lu -ping Department of Pharmacognosy, School of Pharmacy, Second Military Medical University, Shanghai 200433, China Abstract: Objective To explore the anti-anxiety effects of the extract and total valeportriate from Valerianae Rhizoma and the relative mechanism. Methods The behaviors of mice were investigated in terms of using in light/dark transition test, open field test, and the elevated plus maze test and spontaneous activity. The the anti-anxiety effect of the extract and total valeportriate from Valerianae Rhizoma have been studied, serum corticosterone and mouse brain hippocampal tissue in the neurotransmitter 5-serotonin (5-HT), norepinephrine (NE) and dopamine (DA) levels were determined by ELISA kit. Results The behavior manifestations of mice in the light/black transition test, open field test, spontaneous activity, and elevated plus maze experiment of mice were improved by the extract and total valeportriate from Va
