小鼠腹部异位心脏移植模型的建立及改进 陈广顺1，司中洲1，李杰群1，李亭1 中南大学湘雅二医院 普外科 湖南长沙 410011 摘要：目的 探讨稳定的小鼠异位心脏移植模型制作方法，为研究心脏移植排斥反应提供良好的实验工具。方法 选用C57BL/6小鼠做供、受体，采用供体的升主动脉、肺动脉分别与受体的腹主动脉及下腔静脉行端-侧吻合进行同种异基因异位心脏移植。在经典的心脏移植模型基础上，通过供心获取、保存及动静脉切开技术的改进，成功建立了小鼠腹部异位心脏移植模型。结果 预实验80例，成功率为 (6/80) 7.5%。正式实验30例，术后3天存活率达80.0%（24/30）。供体手术时间（14.0±2.5）min，受体手术时间（46.0±8.3）min，其中腹主动脉及下腔静脉阻断时间（18.0±3.5）min，静脉吻合时间（8.0±1.5）min，动脉吻合时间（11.0±2.0）min，成活小鼠受体手术平均失血0.2ml。手术失败的小鼠死亡原因分别为动脉吻合口出血（3例）、动脉吻合口部狭窄1例，静脉吻合血栓形成2例。结论 熟练地血管吻合技术是小鼠异位心脏移植手术成功的关键。 关键词：心脏移植；小鼠 The establishment and improvement of Mouse abdominal heterotopic cardiac transplantation model Chen Guangshun1， SiZhongzhou1, Li Jie Qun 1, Li Ting1 (the second xiangya hospital of central south university hunan changsha 410011,china) 1作者单位：中南大学湘雅二医院，湖南长沙410011 2作者简介：陈广顺（1982-），男，河北廊坊人，普外移植，博士研究生，研究方向：肝脏移植。 Abstract: Objective To investigate the surgical procedures for establish a reliable murine model of heterotopic cardiac transplantation(HCT),with a purpose of providing a excellent experimental tool for the rejective study in HCT.Methods C57bl/6 mice were used as donor and recipient animals respectively.The procedure of HCT in allogenic mice was performed as following:the donor ascending aorta is sutured end-to-side to the recipient abdominal and the donor pulmonary artery is anastomosed end-to-side to the recipient inferior vena cava(IVE)..On the basis of the classic cardiac transplantation, we established the model of HCT successfully through improving certain technical methods.Results We made 80 cases in preliminary experiment,which 6 cases survival Formal experiment of HCT were performed in 30 mice, with a postoperative day(POD) 3 survival rate of 80.0%(24/30).
