丙型肝炎病毒NS5B在Hep3B细胞的表达及活性分析 孔令保， 刘志文， 吴晓玉， 刘好桔， 王园秀 (江西农业大学生物科学与工程学院南昌市发酵应用技术重点实验室 南昌 330045 ) 基金项目：江西农业大学博士基金(2727) Supported by the PhD Foundation of Jiangxi Agricultural University (2727) 作者简介： 孔令保, 男， 江西省南昌市人，博士，美国芝加哥大学医学院博士后， 讲师，主要从事HCV、HBV 等病毒的致病机制和抗病毒药物的研发，发表SCI论文12篇，核心期刊论文3篇。电话：（0791）3813459， E-mail: 摘 要： 目的 丙型肝炎病毒NS5B基因转染人肝癌细胞系Hep3B细胞，检测NS5B的表达与细胞内RNA聚合酶活性。方法 使用脂质体把包含全长丙型肝炎病毒NS5B基因的重组载体pcDNA-5B转染到Hep3B细胞。RT-PCR和Western blot鉴定NS5B基因的表达，荧光素酶试验检测NS5B 的细胞内RNA依赖的RNA聚合酶活性。结果 RT-PCR和Western blot证实转染的NS5B基因在Hep3B细胞成功地进行转录和翻译，荧光素酶试验表明Hep3B细胞表达的NS5B蛋白具有细胞内RNA依赖的RNA聚合酶活性。结论 人肝癌细胞系Hep3B细胞可以成功表达NS5B基因，且表达的NS5B蛋白具备细胞内RNA依赖的RNA聚合酶活性。 关键词： 丙型肝炎病毒； NS5B； Hep3B细胞； 活性 Expression and activity analysis of hepatitis C virus NS5B in Hep3B cells KONG Ling-bao WU Xiao-yu LIU Zhi-wen LIU Hao-ju WANG Yuan-xiu(Nanchang Key Laboratory of Fermentation Application Technology, College of Life Science and Engineering, Jiangxi Agricultural University, Nanchang 330045, China,) Abstract: objective To investigate the expression and activity of transfected hepatitis C virus (HCV) NS5B in human hepatic carcinoma cell line Hep3B and provide a tool for studying the mechanisms of HCV replication and for the identification of NS5B inhibitors.Methods The recombinant vector pcDNA-5B expressing wild-type HCV NS5B gene was transfected to Hep3B cells using Lipofectamine 2000 reagent kit. Expression of the transfected NS5B gene was determined by RT-PCR and Western blot analysis.
