裸小鼠人结直肠癌淋巴道转移模型的建立.doc

稿号：071522 人结肠癌细胞系裸小鼠淋巴结转移的观察 涂经楷1，傅仲学1，叶晓莉2，赵立波3，王春毅1，周建荣1，汪 斌1 （1重庆医科大学附属第一医院普外科，重庆400016；2重庆市西郊医院检验科，重庆400050；3重庆市第三人民医院神经内科，重庆400014） 摘 要： 目的 建立人结肠癌细胞系裸小鼠爪垫淋巴道转移动物模型，并评价卡培他滨抗癌药物效果。方法 裸鼠爪垫皮下注射人结肠癌HCT-116细胞，在1～6周及8周取材，进行HE染色及CEA免疫组化检测，观察种植瘤的成瘤率、生长情况、淋巴结转移规律，并口服卡陪他滨抗癌药物观察其疗效。结果 种植1周后成瘤率100％。肿瘤在3～4周成指数生长。3周时见第一级腘窝腹股沟淋巴结转移(2/5)，到6周时100%转移，其中腹股沟淋巴结转移阳性率76.9%(10/13)；5周时见第二级髂血管旁淋巴结转移（1/5）；到8周时见第三级肾门淋巴结转移（1/5），髂血管旁淋巴结转移（3/5），但均未见肝肺远处转移。淋巴结转移发生在肿瘤指数生长之前，HE染色及CEA免疫组化染色证实淋巴结有弥漫性癌细胞转移浸润。口服化疗药物卡培他滨能有效抑制肿瘤生长及淋巴结转移。结论 成功建立了人结肠癌裸鼠爪垫淋巴道转移模型。此模型快速简便、转移集中、转移率高，且应用此模型能够评价抗癌药物卡培他滨抗癌疗效。 关键词：人结直肠癌 淋巴道转移 裸鼠模型 卡培他滨 中图法分类：R735.3 文献标识码：A Observation of human colon carcinoma cell line Lymphatic Metastasis in nude mice TU Jing-kai1 , FU Zhong-xue1，YE xiao-li2，ZHAO li-bo3，WANG chun-yi1，ZHOU jian-rong1，WANG bing1(1Department of General Surgery,The First Affiliated Hospital of Chongqing Medical University, Chongqing, 400016, 2Test Department, Xi Jiao Hospital Chongqing 400050, 3department of neurology, Third Peoples Hospital Chongqing 400014, China) Abstract: Objective To establish nude mice claw pad model of lymphatic metastasis using human colon carcinoma cell line ,and evaluate anti- cancer medicine curative effect of capecitabine using the model. Methods The lymphatic metastasis nude models were established by injecting colorectum tumor HCT-116 cell line into the nail pad. Collect the tissues in weeks 1-6and 8 after tumor implantation in different parts,observing the formation rate, growing condition and metastasis regularity by HE dyeing and immunohistochemistry stain of CEA,and oral intake of capecitabine observe its medicine curative effect.
