Du ring the p rep aration of samp le s for scann ing electron m icro scop y ( SEM ) , the m ain sou rce of variation in ep iderm al su rface structure is the confu sion of h istological lay ers cau sed by un iden tified stage s in the shedding cycle. U sing a m ethod of gro ss dissection for exam in ing p re served sp eci m en s to determ ine the stage in the shedding cycle, we can identify layers disp en sing w ith h istological techn ique s, and char acterize their surface.
