PAGE 西南林业大学 本科毕业（设计）论文 （2010届） 题 目:干热河谷地区几种豆科植物根瘤菌纯培养及生理生化的初步研究 Preliminary study of several legume rhizobia purification and physiological and biochemical characteristics in dry-hot valley 教学院系: 保护生物学学院 专 业: 植物保护06级 学生姓名: 代志浩 指导教师: 张东华（讲师） 评 阅 人: 刘丽（实验师） 2010年 6 月 干热河谷地区几种豆科植物根瘤 菌纯培养及生理生化的初步研究 代志浩 （西南林业大学 保护生物学学院 昆明 650224） 摘要: 本试验对采自云南元谋和元江干热河谷地区的10种豆科植物根瘤样品进行了分离纯化培养，共纯化出34株菌，其中具有根瘤菌特征的的有4个菌株，对其进行耐酸碱性、耐盐性、生长温度范围3种生理生化的初步分析测定，所测4个菌株均表现出耐酸碱性和耐盐性的性状，在短时间的低温和高温下处理后，仍能保持活性，表现出良好的生长势。并对分离出来的10株革兰氏阴性菌进行回接实验，选择沙土、蛭石、营养液等不同的培养载体，并选择台湾相思作为回接豆科植物，以确定供试豆科植物与所分离培养菌种之间的共生结瘤关系、获得并保存高效优良的根瘤菌菌株。 关键词: 干热河谷 根瘤菌 分离纯化 回接试验 生理生化 Preliminary study of several legume rhizobia purification and physiological and biochemical characteristics in dry-hot valley Dai Zhihao (Faculty of conservation biology ,Southwest Forestry University, Kunming,650 Abstract: This study taken ten kinds of legume rhizobia for purified culture from theYuanmou and Yuanjiang dry-hot valley, and 34 strains were purified, which has the characteristics of Rhizobia strains,4. Determined by the preliminary analysis, 4 strains showed acid and salt tolerance traits, in short low and high temperature treatment, remain active and showing good growth potential.
