12、24、36、48 12、24、36、48 h五个时间点内,800 Irtg/mL浓度的ZF2作用细胞24 h可以最大程 度地促进LDLR、HMGCR、LXR一值、ABCAl mRNA的表达(p0.05),48 h可最 大程度地促进CYP7A1 rnRNA的表达(po.05)。推测张氏马尾藻岩藻聚糖降胆固 醇的机理可能是:促进胆汁酸合成酶CYP7A1 mRNA、肝受体LXR—n mRNA、ABCAl mRNA的表达,增加了胆固醇的转化和排出;影响HMGCR活性,减少细胞内胆固 醇的合成;通过反馈机制激活SREBP一2 mRNA表达,从而诱导其靶基因LDLR表达, 提高细胞对血清中低密度脂蛋白胆固醇的清除能力。由此可见,马尾藻岩藻聚糖降胆 固醇作用是通过多条途径共同实现的且其降胆固醇作用与其多糖浓度、多糖作用时 间、硫酸基含量及单糖组成相关。 关键词:马尾藻,岩藻聚糖,HepG2细胞,胆固醇,机理 Il 万方数据 C C holesterol--lowering Effect and Mechanisms of Sargassum Fucoidan onon HepG2epL/2 1,5ell Abstract TIlis study was conducted to investigate the cholesterol-lowering effect and mechanisms of Sargassum fucoidan,to comprehensive evaluation of the cholester01.10wering activity of six different crude sargassurn fucoidan with chemical and cell experiment in vitro.The crude fucoidan from Sargassum zhangii was isolated and purified by DEAE一52 cellulose chromatograph and determined its chemical composition which cholesterol—lowering activity was best.Discussed the effect and mechanisms on cholesterol metabolism of the different components from Sargassum zhangii fucoidan by HepG2 cell.Finally provide the theory basis for the research on cholesterol-lowering activity of sargassum fucoidan.The main research content is as follows. (1)Comparative studied the cholesterol micellar inhibition rate,bile acid salt binding rate and the effect of the total cholesterol levels on HepG2 cell of crude fucoidan from six different sargassum(Sargassum wightSargassum zhangii,Sargassum hemiphyllum, Sargassum henslowianum.Sargassum naozhouense,Sargassum integerrimum).111e results showed that,crude fucoidan from Sargassum zhangii had a relative advantages of cholesterol-lowering effect which polysaccharide content Was 33.37%,fucose content Was 7.91%and the content ofsulfate Was 13.63%. (2)Using DEAE一52 cellulose chromatograph purified the crude focoidan of Sargassum zhangii,and there were three different components(named ZF 1,ZF2,ZF3).
