The PAS reaction (Periodic Acid-Schiff reaction) is an indispensable histochemical technique used to detect the presence of glycogen, carbohydrates and other components in tissue. Developed in the late 19th century by the German biochemist Dr. Hans Fischer, the technique was further refined by Dr. Remy Martin and Maurice Herzog in the early 20th century.The technique relies on the reaction of periodic acid with glycoproteins and glycogen or other carbohydrate moieties in the tissue being studied. This oxidation reaction, known as the Schiffs reaction, results in a pink or magenta color. Thus, this technique is not a stain per se, but instead a true histochemical reaction.The applications of PAS reaction are varied. It has been adopted for the morphological study of the carbohydrates present in a wide range of tissues, such as bone, cartilage, tendons, and skin. It has also been used in the diagnosis of various diseases, including rheumatoid arthritis, asthma and coronary artery disease. The boundaries of some diseases such as myocardial infarction and stroke can be more easily located by staining tissue sections with PAS based stains.The PAS reaction is also one of the most commonly used histochemical technique in the diagnosis of neoplasms. It is especially useful in identifying metastatic very small carcinoma cells that cannot be detected by routine histological examination. PAS can help differentiate the various types of carcinomas and identify the presence of intracellular glycogen. This can further help in staging and grading the tumor and hence determining the prognosis.In oral histology, PAS is commonly used to assess the development of enamel and dentine in teeth and helps in the evaluation of certain dental diseases. The fungiform papillae of the tongue are identified by the same technique.The PAS reaction is also used in the study of parasites. This is especially useful in the identification of protozoa in fecal smears. It can also help in the
