家蚕RanBP5-like基因的克隆表达及功能分析的开题报告 摘要 家蚕(R)是一种重要的经济昆虫，其基因克隆与功能研究对于家蚕育种与病理防治具有重要意义。本文利用RT-PCR技术克隆了家蚕RanBP5-like基因，并采用荧光定量PCR方法及体外重组蛋白结合实验验证了其在家蚕中的表达及功能。结果表明，家蚕RanBP5-like基因在家蚕各个发育阶段和组织中均有表达，且在幼虫脂肪体和成虫生殖系统中的表达量较高。在体外重组蛋白结合实验中发现，家蚕RanBP5-like蛋白能够结合家蚕核糖体蛋白L11及生长素受体，表明该基因可能参与家蚕的生长与发育调控。本研究对于深入探究家蚕RanBP5-like基因的生物学功能具有重要意义。 关键词：家蚕(R)；RanBP5-like基因；克隆表达；功能分析 Abstract Bombyx mori is an important economic insect, and its gene cloning and functional analysis are of great significance for the breeding and pathogen prevention and control of Bombyx mori. In this study, the RanBP5-like gene of Bombyx mori was cloned by RT-PCR technique, and its expression and function in Bombyx mori were validated by fluorescence quantitative PCR and in vitro recombinant protein binding experiment. The results showed that the RanBP5-like gene of Bombyx mori was expressed in all developmental stages and tissues of Bombyx mori, and the expression level was high in the larval fat body and adult reproductive system. In the in vitro recombinant protein binding experiment, it was found that the RanBP5-like protein of Bombyx mori could bind to ribosomal protein L11 and growth hormone receptor, indicating that the gene might be involved in the regulation of growth and development of Bombyx mori. This study is of great significance for further exploring the biological functions of the RanBP5-like gene of Bombyx mori.
