Abstract
Withtheincreasingprevalenceofmulti-drugresistant(MDR)
bacteria,thedevelopmentofnovelantibioticshasbecomean
urgentpublichealthpriority.Thispapersummarizesthecurrent
statusofnovelantibioticdevelopment,includingtheidentification
ofnewtargets,thediscoveryofnaturalandsyntheticcompounds,
andtheapplicationofinnovativetechnologies.Despitesome
encouragingprogress,therearestillnumerouschallengesahead,
suchasclinicalvalidation,regulatoryapproval,andequitable
access.Nonetheless,thedevelopmentofnovelantibioticsholds
greatpromisefortacklingtheglobalthreatofMDRbacteria.

摘要
随着多重耐药菌的日益普及，新型抗生素的研发已成为迫切的
公共卫生问题。本文总结了新型抗生素研发的当前情况，包括
发现新的靶标、发现天然合成化合物以及应用创新技术。尽管
已经取得了一些有益的进展，但仍然存在许多挑战，例如临床
验证、监管批准和平等获得权等。然而，开发新型抗生素对于
解决多重耐药性细菌的全球威胁充满希望。

Introduction
Sincethediscoveryofpenicillinin1928,antibioticshave
revolutionizedthepracticeofmedicineandsavedcountlesslives.
However,theoveruseandmisuseofantibioticshaveledtothe
emergenceandspreadofmultidrug-resistant(MDR)bacteria,
whichposeaseriousthreattoglobalhealth.Accordingtothe
WorldHealthOrganization(WHO),antibioticresistanceisoneof
thetoptenglobalpublichealththreatsfacinghumanity.Inrecent
years,therehasbeenagrowingrecognitionoftheurgentneedfor
thedevelopmentofnovelantibioticstocombatMDRbacteria.
IdentificationofNewTargets
Thetraditionalapproachtodrugdiscoveryhasreliedonscreening
chemicallibrariesforcompoundsthatcaninhibitthegrowthor
replicationofbacteria.However,thismethodhasencountered
diminishingreturns,asmanypotentialdrugtargetshavealready
beenexploited,andtheeffectivenessofexistingantibioticshas
decreasedduetores
