  锌指蛋白 Zfx 在人脑肿瘤干细胞中的表达与 意义# 饶竞，赵洪洋，杨海峰，黄启伟，杨林，姚东晓** 5 10 15 20 25 30 35 40 （华中科技大学同济医学院附属协和医院，武汉 430022） 摘要：X 染色体耦联锌指蛋白（Zfx）是锌指蛋白超家族中的一员，是胚胎干细胞和成体干 细胞自我更新的共同分子基础，Zfx 在人脑肿瘤干细胞(BTSCs)中的表达有待探讨。本研究 从人脑胶质瘤细胞系 U251 和 U87 中分选和鉴定脑肿瘤干细胞，通过免疫蛋白印迹法对样本 中 Zfx 的表达进行检测，应用统计学方法对人脑肿瘤干细胞中 Zfx 的表达进行分析。研究发 现 CD133 阳性细胞在人脑胶质瘤中所占比例极其微小：U251 约 1.97% ± 0.53%，U87 约 1.32% ± 0.34%。CD133 阳性细胞在无血清培养基中可以形成肿瘤球，且神经干细胞标志 物 nestin 染色阳性；未分化肿瘤球在完全培养基中分化为肿瘤细胞，并表达与来源胶质瘤细 胞相似的神经元标志物β-tubulin 和星形细胞标志物 GFAP。未分化肿瘤球中 Zfx 的蛋白表 达水平明显高于其在分化肿瘤球和来源胶质瘤细胞中的表达。Zfx 的蛋白表达在人脑肿瘤干 细胞中异常增高，它有可能是脑肿瘤干细胞自我更新、抗凋亡的一个关键性的调控因子。 关键词：胶质瘤；脑肿瘤干细胞；X 染色体耦联锌指蛋白（Zfx） 中图分类号：R739.41 The expression of zinc finger protein X-linked in brain tumor stem cells of human glioma Rao Jing, Zhao Hongyang, Yang Haifeng, Huang Qiwei, Yang lin, Yao Dongxiao (Union Hospital, Tongji Medical College, Huazhong University of Science and Technology, Wuhan 430022) Abstract: Zinc finger protein X-linked (Zfx) is a zinc finger protein of the Zfy family. As a shared transcriptional regulator of embryonic and adult stem cells, Zfx may be a common molecular basis of stem cells self-renewal. In this study, we investigated the expressions of zinc finger protein X-linked (Zfx) in brain tumor stem cells (BTSCs) of human glioma.We isolated and identified brain tumor stem cells from two glioma cell lines U251 and U87. The expressions of Zfx were measured by western blot in protein level and analyzed by statistics.The ratio of CD133-positive cells of U251 cells and U87 cells was 1.97% ± 0.53%; 1.32% ± 0.34%. The isolated CD133-positive cells formed tumor spheres in serum-free medium. Undifferentiated spheres from both U251 and U87 were immunostained for characteristic neural stem cell marker nestin. Under differentiation conditions, the tumor spheres were differentiated in complete medium and expressed neuronal marker β-tubulin and astrocytic marker GFAP, similar to those in the original glioma cell lines.
