南开大学硕士研究生毕业(学位)论文 Abstract ABSTRACT Cancerhavegreatlythreatenedthelifeofhumanbeings.Traditional chemotherapeuticdrugshavedeficienciesinclinicalusebecausetheykillednormal cellswhileinhibitingtumorcells.Endogenielowmolecularweightanti-canceragents canselectivelyinhibitmanykindsofcancercellsandhavenocytotoxcitytonormal humanceps.Becauseofitslimitedresnutce,We-studiedthecherniealstrucluirand 1htanticancermechanismoftheselowmolecularweightagents,inordertodevelop newandsafechemotherapeaticdurgs,atthesametimeprovidingthetheoryfor discoveringtheoriginofcancer. Throughion-exchangechrcimatograph,MTTreductiontestandaminoacids analysis,wefound3effectiveedrnpounds,whichwereTaurine.Ornithine.Camosine 加menmponentsemuence-Sand1. Wt:studiedtheeffectofdifferentconcentrationofTaurine.Ornithine.Camosine ontheproliferationofcancercellsHL-60,comparedthemwiththenormalhuman lymphaticcells.Inordertoinvestigatethemechanismoftheirinhibitiononcancer cells,westudiedthefragmentofnuclearDNA,theacitivityofoxygenfreeradical andthelevelofp45/skp2andppk/7.i1 Experimentalresultsshowedthat:these〕 compoundscanselectivelykillHL-60cellsalvwlvwdegree,whilehavingnoeffect tonormallymphaticcells:ThenecessaryaminoacidsinsampleLWM extraction.effluence.5and7showedsynergeticeffectwhenmixturedwith Taurine,OrnithineandCamosine.TheycandegradenuclearDNAofHL-60cellsinto 3okbrfagment,orDNAladder(causedbyhighconcentrationCamosine),through whichHL-60cellsenteredapoptosis.AfterHL-60cellswasincubatedwiththethree drugs,theaeitivityofoxygenfreeradicalwasgreatlydecreased,whichindicatedthal oxygenthreateningmay6t=1brinductionofapoptosis.Immunuhistologicalstaining showedthatupregulatingthelevelofp45/skp2anddownregulatingthatofp27/kipare alsocorrelatedwithapoptosismechanism inducedbyTaurine,Ornithineand Camosine.
