Experimental Study on Effect of Trametes Robiniophila Extract Independently or in Combination with Rh-Endostain and DDP to Inhibit Tumors Zhou Meixiang, Sun Xiuhua (author) Zhao Danyi, Liu Fang, Sun Jinghua, Zhang Yang Tumor Department, No.2 Affiliated Hospital of Dalian Medical University, 116027 Abstract: Purpose: To investigate the growth inhibition and apoptosis inducement effect of trametes robiniophila murr(PS-T) combined with rh-Endostain or with cisplatin in vitro on human lung adenocarcinoma cell line A549.Method: Culturing exponential phase human lung adenocarcinoma cell line A549 with PS-T in different concentrations and culture solution of PS-T, Rh-Endostain, Cisplatin for 24h, 36h, or 48h. Then apply MTT colorimetry to test OD, calculate the inhabitation concentration, and at the same time test the apoptosis rate result of flow cytometry. All the data are expressed as mean± standard deviation, then using statistical package SPSS13.0 to deal with it refers to statistics. At last, one-way analysis of variance should be adopted to find the discrepancy of Trametes Robiniophila Murr in different concentration, and are adopted to test and analysis the discrepancy with two different combinations in different groups. When P 0.05, it means that the discrepancy makes sense in statistics. Result:1. PS-T significantly has inhibitation effect on human lung adenocarcino- ma cell line A549. Its inhabitation concentrations increases as the concentration goes up, and the inhibitation concentrations is the highest when concentration comes to 16mg/ml. Among the three different time scale, 48h can produce the best result. The flow cytometry can test that apoptosis inducement rate of trametes robiniophila murr on human lung aden- ocarcinoma cell line A549 also increases when the concentration goes up.2.
