To study an available method of antigen retrieval is a goal to achieve both good mi munochem istry result and preserving retinal proteins. Objective The ami of present study is to investigate tyrosin digestion, hightemperature heat pressure, w ater bath heating and microw ave repair in antigen retrieval for retinal tissues. Methods Retinal tissue was isolated and obtained from clean Chinchilla rabbits. Four hundreds retinal paraffin sections were prepared. Four kinds of antigen retrieval methods for retinal tissue including tyrosin digestion, hightemperature heat pressure, w ater bath heating and m icrowave repair w ere used respectively. The depigmentated retinal paraffin sectionw ithout antigen retrievalw as used as contro.l The positive rates of expression of CRAL P, Rhodopsin and opsin proteins w ere evaluated and compared among four kinds of antigen retrieval methods by mi munochem istry. Results CRAL P, Rhodopsin and opsin proteinwas positively expressed in cytoplasm of retinal pigment epithelial cells and the outer segment of photoreceptor respectively.
