维普资讯 ChineseJournalofBiomedicalEngineering(EnglishEdition)Volume16Number4，December2007 TheSynergisticEffectofHT andComplement RegulatoryProteinsin ResistingtheImmunological LISheng-zhi，LIU Bing-qian，W ANG Guang,you， ZHANG Yue，M A Zhi-fang，MA Teng-xiang TianjinInstituteofUrologicSurgery，Tianjin300211，China Abstract．Objective：ToestablishthepolytransgenicmiceexpressingthehumanHT andcomplementregulatoryproteins(CRPs)anddiscusstheirabilitytoresistthehyper— acuterejection(HAR)anddelayedxenograftrejection(DXR)ofhetemgenictransplanta— tion．Methods：Transgenicmicewereproducedbymicroinjectiontoconstructgeneforhu— mallHT，delayacceleration~cotr(DAF)and／orCD59intohtemalepronucleusofzy— gote．PCR andSouthernblotwereusedtoscreenthepositivetrarisgenicmice．Flow cv． tometry(FCM)wasusedtodetecthteexpressionofHT，ot—GalandDAForCD59onhte PBMCsoftransgenicmice．Thesurvivaltimeandfunctionofhteheartoftransgenicmice weredetermined by amodified Langendorff cardiacperfusion appar~itus：Th echange of proteinosison IgM、IgG、C3candC9 from differentcardiacvascular：endothelialcellsof transgenicmiceweredetectedbyimmunohisotchemistry．Results：HT，DAForCD59were highlyexpressedonhtepositivetransgenicmicebyFCM．ThedepositionofIgM、IgG、C3c orC9inhtecardiacvascularendohtelialcellsofhtepositivetransgenicmicewerede— creased．Th esurvivaltimeandfunctionofhteheartofhteco—transgenicmicewithABser— um perfusionweresignificantlylongerandhigherthanthatofhtesingleHTpositivetrans— genicmice(P0．05)．Conclusion：Themiceco—expressingHT／DAForHT／
