Methods: Hu- man mast ce s were dispersed from co on tissue w ith co agenase and hya uronidase, and w ere cha enged w ith stimu u s for 15 min at 37e . Tryptase assay performed fo ow ing previous procedures . In brief , a 96-w e microtitre p ate w as coated w ith antiserum to human tryptase. T he tryptase eve s in the samp es were detected w ith a monoc ona antibody specif ic to tryptase and the reaction w as visua ized by addition of OPD. Resu ts: At 15 min and 35 min fo ow ing incubation, ant-i IgE and ca cium ionophore were ab e to provoke signif icant tryptase re ease from hu- man co on mast ce s . Chymase inhibitors ZIGPFM , TPCK and A- antitrypsin had no stimu atory eff ect on co on mast ce s at both 15 min and 35 1 min incubation periods .
