响应曲面法优化亚麻籽蛋白提取工艺

2. 青海 西宁 81000pH、时间、料液比对亚麻籽蛋白提取率的影响,并建立该工艺的二次项模型。结果表明,回归模型具有高度显著性,可以对亚麻籽蛋白的提取率进行很好的分析和预测,并最终确定亚麻籽蛋白最佳工艺条件为:料液比1:2050℃,浸提液pH值为10,在此条件下的亚麻蛋白提取率为79.26%,纯度达到92.34%。

Optimization of extraction processing for flaxseed protein
by response surface methodology

Abstract: Alkali-solution and acid-isolation was used to extract flaxseed protein from flaxseed in this study. Based on singer factor test, response surface method was used to study effects of extraction temperature, extraction pH, extraction time and solid-liquid ratio on extraction rate of flaxseed protein, and a second order quadratic equation for flaxseed protein extraction was finally obtained. The results showed that the model was of great difference, and the extraction of flaxseed protein from flaxseed was analyzed and predicted by the model. The optimum extraction of flaxseed protein from flaxseed were solid/liquid ratio(g/mL)1:20, extraction time 90 min, extraction temperature 50℃, extraction pH 10.0, the extraction rate of flaxseed protein from flaxseed was 79.26% and purity reached 92.34% under the optimal conditions.

亚麻(Linum usitatissimum L.)[1],主要分布在北美、印度、中国等国家,是世界上最古老的纤维作物,品种较多,已成为世界十大油料作物之一[2-4]。亚麻籽(flaxseed 或linseed)是亚麻的种子,含有油脂和蛋白质等营养成分。其中,脂肪占31.9~37.8%21.9~31.6%[2];另外,亚麻籽中还含有一定量的亚麻籽胶,随品种和栽培区域不同而不同,约占种子质量的2~10%[]。诸多文献[6-9]研究表明,油料蛋白质的提取主要有等电点沉淀、离子交换、碱溶酸沉法、反胶束溶液萃取、膜分离、酶法等方法,其中碱溶酸沉法是我国普遍采用的蛋白质提取工艺,该方法能够有效地提高蛋白质的提取率,充分利用蛋白质资源。
