To some extent, the current marketed macrolides are limited for clinical application. With the increasing incidence of drug- resistant bacteria, its urgent to develop some new macrolide antibiotics to combat with these bacteria. These years, researchers focus on improving pharmaceutical properties of high active compounds and changing dosage form in order to enhance macrolide antibiotics bioavailability. This article fi rst introduces a new type of macrolide—ketolide, then make a brief introduction of telithromycin which was approved in 2001, and reviews briefl y the advance in the investigational macrolide antibiotics within this decade, among which kelimycin, cethromycin, solithromycin and modithromycin are the main drug candidates.
