In the exper iment, xy nA w as amplified from the genome of Ba cillus p umilus and cloned into E. coli and B. subtilis Shuttle Vector GJ148, resistance gene of specti nom cin w as also cloned into it . A signal peptide ( Ep r) w as cloned into the constructed scr eening vec tor pYG for the expression of xy nA gene in B acillus subtilis WB700. T he screening vector pYG w as successful constructed, and the signal peptide ( Ep r) successful mediated the expression of xy nA gene in Bacillus subtilis WB700. T his experiment show ed the possibilit of constructing a screening vector for all signal peptides fr om B acillus p umilus , it could successful express x lanase gene w ith cloned signal peptide.
