In order to identify whether it happens natural mutation or due to mixed with hybrid seed, we collect and cultivate 9 samples of the same kind of seed coat in red with the target. Extracting DNA and using SSR molecular marker technology detect the nine varieties species with normal and early heading plant. It turned out that, using nearly 50 pairs of SSR primers to identify the genetic relationship between the target plant and 9 varieties, through DNA extraction, PCR amplification and agarose gel electrophoresis process, we compared and analyzed with the result of the amplification. There are differences between the 9 samples and the normal and early heading foxtail millet, but the normal and early heading millet are nearly the same. These results suggest that the mutant of foxtail millet we found is coming from the natural mutation in Tong gu, rather than mixed with the hybrid seed.
