* This procedure applies to the clinical laboratory environment with the BD FACSCount flow cytometer for the purpose of CD4 lymphocyte enumeration using whole blood specimens. We will use this procedure during the laboratory exercise Prime the system Disconnect both sheath tanlk and waste tank by removing quik-disconnects and sensors for both tanks. Empy the waste, add 200 ml hypochlorite Fill the sheath tank with sheath fluid up to one inch from the top. Replace reserviors, fluidic tubing and sensors. Check for bubbles in both the fluid filter and in the flow cell. Enter control reagent lot information using the keypad. Control lot information can be found on the control bead tray. Reagent lot information can be found on the foil bag. Run the CD4 tube before running the CD8 tube. Always vortex tubes for 5 seconds upright before pressing RUN on the FACSCount Console. Record QC data using SOP 1006 as a daily form for running controls.
