柚皮苷脂质体缓释凝胶制备工艺优化 摘要:考察影响柚皮苷脂质体缓释凝胶制备工艺的各因素,以包封率为指标,确定柚皮苷脂质体缓释凝胶最佳制备工艺条件。以成型性、涂展性、稳定性的综合作用为指标,采用正交试验设计对影响凝胶剂质量的卡波姆940用量、载药量、5%羟苯乙酯乙醇溶液和三乙醇胺用量进行考察,并用高效液相色谱法测定所制凝胶中柚皮苷的含量。确定柚皮苷脂质体缓释凝胶最佳制备方法为以乳化-低温固化法制备柚皮苷脂质体;柚皮苷脂质体缓释凝胶处方为0.3 g卡波姆940为凝胶基质、2 g 10%月桂氮卓酮无水乙醇溶液为促渗剂、1.0 g三乙醇胺为pH调整剂、0.4 g甘油为保湿剂、0.4 g 5%羟苯乙酯乙醇溶液为防腐剂、载药量为2.08%。柚皮苷脂质体缓释凝胶剂制备工艺简单,所制凝胶质地均匀细腻,稳定性良好。 关键词:柚皮苷;脂质体;缓释凝胶;正交试验设计 中图分类号:R282 文献标识码:A 文章编号:0439-8114(2016)05-1252-04 DOI:10.14088/j.cnki.issn0439-8114.2016.05.041 Optimization of the Preparation Process of Naringin Liposome Sustained-release Gel WANG Hai-yan,CHEN Yue-hua,LI Juan,CHEN Qiong (Xinyang Engineering Research Center for the Exploitation of Chinese Medicine Resources/Xinyang Key Laboratory of Detection and Analysis of Chinese Medicine/Xinyang College of Agriculture and Forestry, Xinyang 464000,Henan,China) Abstract: The factors which affect the preparation of naringin liposome sustained-release gel and establish the optimal preparation process were studied. The preparation method of naringin liposome was investigated with encapsulation percentage as selection standard. Formability, spread performance, stability were used as comprehensive evaluation indicator to select the preparation process by orthogonal design, taking the dosage of carbopol 940, drug loading, 5% hydroxy ethyl benzene ethanol solution and triethanolamine as factors and the content of naringin in gel was determined by HPLC. The naringin liposomes were preparaed by the method of emulsion-low temperature curing. The prescription of naringin liposome sustained-release gel was as follows:substrat of carbopol 940 was 0.3 g, penetration enhancers of 10% azone ethanol solution was 2 g, pH regulator of triethanolamine was 1.0 g, moisturizer of glycerol was 0.4 g, preservatives of 5% hydroxy ethyl ethanol solution was 0.4 g, the drug loading was 2.08 g naringin per 1 g of the gel.
