不同剂量三氧化二砷单独及联合顺铂对人宫颈癌移植瘤的作用及机制[摘要] 目的 探讨不同浓度的三氧化二砷（As2O3）与顺铂单独或合用对宫颈癌裸鼠移植瘤的疗效和毒副作用。 方法 取人宫颈癌细胞接种于裸鼠的皮下，建立裸鼠移植瘤模型，按照用药的不同分为观察组六组与对照组一组，对照组给予生理盐水用药；观察组六组分别给予三氧化二砷单独使用、顺铂单独使用及顺铂联合三氧化二砷使用；通过流式细胞仪检测细胞凋亡率、HE染色观察用药后的毒副作用。 结果 观察组各组的瘤质量及细胞的凋亡率显著优于对照组，结果具有统计学差异（P0.05）。观察组各组药物治疗后出现一定程度上的白细胞降低，顺铂药物组出现骨髓抑制明显；其他血红蛋白、血小板值较对照组显著下降；用药后观察组裸鼠的血尿素氨水平、丙转氨酶值有所升高；顺铂药物组的肝肾损伤较为明显，与对照组相比数值显著升高。 结论 三氧化二砷联合顺铂治疗宫颈癌移植瘤，有效的促进了癌细胞的凋亡，增强了抑制癌细胞增殖的作用，促进了临床的治疗效果，临床效果显著，值得推广。[关键词] 宫颈癌移植瘤；三氧化二砷；顺铂；裸鼠[中图分类号] R737.3 [文献标识码] A [文章编号] 2095-0616（2015）01-44-04[Abstract] Objective To study the effect and toxicity of different concentrations of arsenic trioxide （As2O3） and cisplatin used alone or in combination on cervical cancer in nude mice. Methods Human cervical cancer cells were inoculated subcutaneously in nude mice， nude mouse xenograft modelas were divided into six observation groups and one control group according to the different treatment ，the control group was given normal saline treatment.The observation group of six groups were treated with arsenic trioxide alone， cisplatin alone and combined use of two.To detect apoptosis by flow cytometry， and the toxic side effect was observed by HE staining. Results Tumor mass and apoptosis rate of cell in observation group were significantly better than the control group， P0.05.After the treatment，observation group appeared to blood cell decrease ， and cisplatin group appeared bone marrow suppression significantly.Hemoglobin， platelet values significantly lower than control group.After treatment， the nude mice blood urea nitrogen level， alanine aminotransferase values increased.Liver and kidney injury in cisplatin group increased obviously than the control group. Conclusion Arsenic trioxide combined with cisplatin in the treatment of cervical cancer xenograft promote the apoptosis of cancer cells and the clinical therapeutic effect effectively， enhanced the inhibition of cancer cell proliferation， it is worthy of promotion with good clinical effect.[Key words
