水磨年糕中微生物的分离纯化与鉴定 　　摘 要：通过对年糕内部和表面的微生物进行分离纯化，并通过形态观察、生化测定的方法来鉴定微生物的组成，结果表明：年糕表面的微生物主要有6种细菌、3种霉菌和2种酵母菌。细菌中4种是芽孢杆菌，分别为巨大芽孢杆菌（Bacillus megaterium）、枯草芽孢杆菌（Bacillus subtilis）、蜡状芽孢杆菌（Bacillus cereus）和短芽孢杆菌（Bacillus brevis），2种是非芽孢杆菌，分别为乙酰短杆菌（Brevibacterium acetylicum）和乙酸钙不动杆菌（Acinetobacter calcoaceticus）；霉菌分别为米根霉（Rhizopus oryzae）、构巢曲霉（Aspergillus nidulans）和米曲霉（Aspergillus oryzae）；酵母菌分别为茁芽丝孢酵母（Trichosporon pullulans）和白色假丝酵母（Candida albicans）。年糕内部的微生物有巨大芽孢杆菌、枯草芽孢杆菌、短芽孢杆菌和白色假丝酵母，无霉菌。 　　关键词：年糕；分离纯化；微生物；鉴定 　　中图分类号 R155 文献标识码 A 文章编号 1007-7731（2015）19-25-04 　　Purification and Identification of Microbes in Water Mill Rice Cake 　　Chen Ting1，2 et al. 　　（1School of Marine Sciences，Ningbo University，Ningbo 311300，China；2Jiangbei Science and Technology Bureau，Ningbo 311300，China） 　　Abstract：Through the morphology observation and the physiological and biochemical of microorganisms which were separated and purified from the interior and the surface of the rice cake，we can get that there were 6 kinds of bacterium（involving 4 kinds of Bacillus which were Bacillus megaterium，Bacillus subtilis，Bacillus cereus and Bacillus brevis，2 kinds of non-Bacillus were Brevibacterium acetylicum and Acinetobacter calcoaceticus），3 kinds of molds（Rhizopus oryzae，Aspergillus oryzae and Aspergillus nidulans），and 2 kinds of Yeast（they were respectively Candida albicans and Trichosporon pullulans）on the surface of rice cake.The inside microorganisms of Rice cake were Bacillus megaterium，Bacillus subtilis，Bacillus brevis and trichosporon pullulans without mold. 　　Key words：Rice cakes；Separation and purification；Microbes；Identification 　　年糕是一种历史悠久且地方特色非常浓郁的方便性米制品[1-2]，水分含量高达45%，营养丰富。年糕在其生产过程中极易受到微生物的污染，其原辅料、加工半成品及成品都直接暴露于空气和水中，在成熟后期的切片、晾晒和装袋的过程中很容易受到空气中的微生物的二次污染[3]。因此，即使是真空包装的年糕也会因一些细菌芽孢的存在而发生腐败。本文通过对年糕表面和内部的微生物进行分离纯化及鉴定，并对年糕表面与内部的微生物进行分析比较，探索提高年糕保质期的方法，为企业开发适合年糕的保鲜措施提供依据。 　　1 材料与方法 　　1.1 材料与仪器 　　1.1.1 材料与试剂 年糕：宁波市江北慈城庄永大年糕厂提供。蛋白胨：BR，临安木木生物技术有限公司；琼脂：AR，临安木木生物技术有限公司；牛肉膏：BR，杭州百思生物技术有限公司；氯化钠：AR，西陇化工股
