吉西他滨、顺铂及香菇多糖对Lewis肺癌细胞株抑制作用的实验研究 　　[摘要] 目的 研究吉西他滨、顺铂、香菇多糖三种药物单独使用及联合使用对Lewis肺癌细胞株的增殖抑制作用。 方法 分别使用不同浓度的吉西他滨（0.5、1、2、5、8、10、12 μg/mL）、顺铂（1、2、4、8、10、16、20 μg/mL）、香菇多糖（50、100、150、200、250、300、400 μg/mL）单独及联合处理Lewis肺癌细胞株，24 h后MTT法测定细胞生长抑制作用，应用金氏公式进行联合用药分析。 结果 吉西他滨、顺铂、香菇多糖对Lewis肺癌细胞均有不同程度的增殖抑制作用，两种或三种药物联用后对Lewis肺癌细胞株的抑制率显著增加，其中8 μg/mL的吉西他滨与10 μg/mL的顺铂和250 μg/mL香菇多糖联用时，其抑制率近似于10 μg/mL的吉西他滨和16 μg/mL的顺铂两药联用；而当香菇多糖的浓度增加至200 μg/mL时，三药联用的抑制率明显高于吉西他滨和顺铂两组药物联用（P 0.05）。 结论 香菇多糖可增强吉西他滨和顺铂对Lewis肺癌细胞的增殖抑制作用，表现为协同效应，并且联用后能够减少常规化疗药物的用量。 　　[关键词] 吉西他滨；顺铂；香菇多糖；Lewis肺癌细胞株；MTT法 　　[中图分类号] R734.2 [文献标识码] A [文章编号] 1673-7210（2016）09（b）-0018-04 　　[Abstract] Objective To investigate the anti-proliferative effect of single use of Gemcitabine， Cisplatin， Lentinan and combination of them on Lewis lung cancer cell lines. Methods Lewis lung cancer lines were treated with different concentrations of Gemcitabine （0.5， 1， 2， 5， 8， 10， 12 μg/mL）， Cisplatin （1， 2， 4， 8， 10， 16， 20 μg/mL）， Lentinan （50， 100， 150， 200， 250， 300， 400 μg/mL） and combination of them separately. After 24 hours， the inhibitory effects were measured with MTT assay and Jin formula was applied to analyze the combination effect of the drugs. Results Gemcitabine， Cisplatin and Lentinan all showed the anti-proliferative effects on Lewis cancer cell lines in varying degree. The inhibitory rates of any two or three drugs of them were significantly higher， among which， the inhibitory rate of combination use of 8 μg/mL Gemcitabine， 10 μg/mL Cisplatin and 250 μg/mL Lentinan was close to that of 10 μg/mL Gemcitabine and 16 μg/mL Cisplatin. When the concentration of Lentinan increased to 200 μg/mL， the suppression rate of the tumor in three-drug regimen group was higher than that in the Gemcitabine combined with Cisplatin group （P 0.05）. Conclusion Gemcitabine， Cisplatin shows the anti-proliferative effect on Lewis lung cancer cell lines distinctly， and the drugs have better cooperative effect combined with Lentinan， further more， th
