[作者简介]谭茂兰，女，硕士研究生，主要从事中药炮制学研究；通讯作者：黄勤挽，男， 博士，副教授，主要从事中药炮制学研究。 [基金来源]国家自然科学基金，四川省科技厅支撑计划（2013SZ0140），四川省 中医药管理局青年基金（2010-12）。 附子蒸制过程中酯型生物碱的含量变化规律研究 谭茂兰，黄勤挽*，易佳佳，范润勇，韩丽 （成都中医药大学，成都 611137） [摘要]目的：研究鲜附子在常压蒸制过程中 6种酯型生物碱的含量随蒸制时 间延长的变化，探讨蒸制过程中酯型生物碱变化规律，为蒸制加工附子提供依据。 方法：采用 Phenomenex Gemini 色谱柱（250×4.6mm，5μm），以 0.04 mol?L-1 乙酸铵（用氨水调 pH 至 9）为流动相 A，乙腈为流动相 B，梯度洗脱，测定各样 品的酯型生物碱含量。 结果：3种单酯型生物碱随蒸制时间的延长呈 M型变化， 3 中双酯型生物碱随蒸制时间含量逐渐降低；在 1h 蒸制时间内单酯型生物碱含 量随双酯型生物碱的减少而增加，在 1～2h双酯型生物碱的含量减少到不能检出， 单酯型生物碱含量有所减小。2～4 小时单酯型生物碱含量逐渐增加并在 3～4h 含量达到最高，随后随蒸制时间的延长而减少。结论：提示鲜附子蒸制 3～4h 可能是较好时间条件即可降低毒性，又保存药效；单酯型生物碱在 1～2h 内减少 但在 2～4h含量增加的原因需进一步研究。 [关键词]附子；蒸制；酯型生物碱；含量变化 Study on The Variation of Ester-type Alkaloids Content in Radix Aconite Lateralis at Different Steaming Time TAN mao-lan , HUANG qin-wan* , YI jia-jia ,FAN run-yong , HAN li (Chengdu University of Traditional Chinese Medicine,Chengdu 611137,China) ［Abstract］Objective: Study on the variation of six ester-type alkaloids content in Radix Aconite Lateralis with the changes of steaming time; Discusses the changes rμles of ester-type alkaloids in the process of steamed and provide the basis for processing Radix Aconite Lateralis by steaming. Method: HPLC analysis was performed on a Phenomenex Gemini (4.6 mm×250 mm,5μm) with 0. 04mol/L ammonium ace-tate (adjust PH value to nine with ammonia，phase A) and acetonitrile (phase B) as mobile phase. Gradient elution was used to the determination the content of ester-type alkaloids in crude aconite roots at different steaming time. Resμlts: The contents of three kinds of monoester-type alkaloids showed a type of M change while three diester-type alkaloids decreased gradually with different steaming time.The contents of monoester-type alkaloids gradually increased with the decreasing of diester-type alkaloids in 1 hour steaming time.
