齐齐哈尔大学毕业设计(论文) PAGE 26  PAGE 26  PAGE \* MERGEFORMAT 0 (此文档为word格式，下载后您可任意编辑修改！) 齐 齐 哈 尔 大 学 毕业设计（论文） 题 目 香豆素衍生物荧光分子探针的合成及性能研究 学 院 化学与化学工程学院 专业班级 学生姓名 指导教师 成 绩 2012年6月12日 摘　要 以香豆素-3-甲酸为原料，经氯乙酰化得中间体香豆素-3-甲酰氯，进一步与2-吡啶-8-氨基喹啉反应，得荧光分子探针(SHK-1)；香豆素-3-甲酰氯与过量的乙二胺反应合成得到SHK-2；2-吡啶-8-氨基喹啉经氯乙酰化得中间体，进一步与无水哌嗪反应，最后与香豆素-3-甲酰氯合成得SHK-3。上述产物的结构经1H-NMR表征，均为目标产物。 经初步筛选后，研究了荧光分子探针SHK-1在中性缓冲溶液中对金属离子的识别性能。结果表明，SHK-1可以选择性荧光猝灭识别Cu2+、Fe3+。SHK-1与Fe3+络合后，SHK-1在波长为351nm处，荧光峰强度猝灭31%；与Cu2+络合后，SHK-1在474nm处，荧光峰强度几乎完全猝灭，猝灭比率达97%。定量分析显示，SHK-1与Cu2+形成了1:1型络合物。金属离子竞争实验表明，常见的金属离子均不干扰SHK-1对Cu2+的检测。 关键词：香豆素；荧光分子探针；Cu2+ Abstract Coumarin-3-methyl chloride was synthesized using the raw marterial coumarin-3-formic acid through chloroacetylation. which were used as mediums of synthesizing complicated fluorescent probes. Furtherly, used coumarin-3-methyl chloride and 2-pyridine-8-aminoquinoline, fluorescent molecular probes SHK-1 was designed and synthesized through reaction. SHK-2 was synthesized using the coumarin-3-methyl chloride and excess ethylenediamine.. 2-pyridine-8-aminoquinoline was chloroacetylated using the raw marterial , which is the mediums of synthesizing complicated fluorescent probes. And then further reaction with anhydrous piperazine, finally the SHK-3 was synthesized through coumarin-3-methyl chloride. The structure was confirmed by 1H-NMR characterization of the above products, are the target product. After initial screening, fluorescent molecular probes of the SHK-1 was studied in the recognition performance of the metal ions in the neutral buffer solution.
