A new source of marine compounds D19 anthracycline anti-tumor activity and its mechanism

PAGE \* MERGEFORMAT 15 A new source of marine compounds D19 anthracycline anti-tumor activity and its mechanism Of: Yuan Jie, He Zhenjian, Wu Jue-heng, Zhu Xun [Abstract] Objective To investigate the novel marine-derived compounds D19 anthracycline anti-tumor effects and molecular mechanisms. Methods tetramethyl nitrous tetrazolium (MTT) assay, terminal deoxynucleotidyl transferase mediated nick end labeling method (TUNEL), Western blot (Western Blotting) D19 detected in human breast cancer cell line MCF-7 and MDA-MB-435 growth and the related signal pathway proteins. Results D19 on MCF-7, MDA-MB -435 half inhibition concentration (IC50) were 4.01 mol / L, and 7.32 mol / L; D19 has effectively induced the apoptosis of human breast cells; Western Blotting showed that the cells of D19 role of apoptosis proteins such as caspase 9 and the downstream effector protein PARP cleavage occur. Conclusions from the cellular level and molecular level prove that the new marine-derived compound D19 anthracycline anti-cancer activity has a strong, may develop into breast cancer and other solid tumors of the lead compound . [Keywords:] marine symbiotic microbes; anti-tumor; anthracycline compounds; human breast cancer cells; apoptosis Abstract: ObjectiveTo study the antineoplastic activity and mechanism of D19, a novel anthracycline derived from marine commensal microorganisms at both cellular level and molecular level. Methods The inhibitory effects of D19 on growth of breast cancer cell lines were determined by MTT assay. The molecular signaling pathways of apoptosis were measured by terminal deoxynucleotidyl transferase (TDT)-mediated biotinylated deoxyuridine-triphosphate (dUTP)-biotin nick end labeling (TUNEL) staining and western blotting. Results IC50 of D19 on MCF-7 and MDA-MB-435 cell lines were 4.01 mol / L and 7.32 mol / L respectively. TUNEL staining showed that D19 could induce apoptosis in MCF-7 cells. In addition, Western blot detected some apoptosis
