PAGE \* MERGEFORMAT 12 Metallothionein gene transfected 1A cells in rat hepatic stellate [Abstract] Objective: To observe the metallothionein gene 1A (MT1A) on hepatic stellate cells (HSCs) proliferation. METHODS: MT1A gene eukaryotic expression plasmid transfected HSCs. By RT PCR, Western blotting detected import HSCs expression plasmid, using AgNOR staining and Ki 67 antigen immunocytochemical staining HSCs proliferation index. Results: MT1A gene over-expression in HSCs transfected MT1A Genome AgNOR number of particles per cell (5.43 + -0.45) compared with pcDNA3.1 group (3.94 + -0.39) and non-transfected group (4.14 + -0.34) were higher (P lt;0.05), immunocytochemistry showed that cells transfected MT1A gene stained positive for Ki 67, the control group was negative. Conclusion: Exogenous MT1A overexpressed gene in HSCs can promote their proliferation. [Keywords:] hepatic stellate cells, metallothionein, proliferation, liver fibrosis, rat Metallothionein (metallothionein, MT) are a class of low molecular weight (6000 ~ 7000), cysteine-rich metal-binding proteins, including MT1, MT2, MT3, MT4 4 subtypes, which is MT1A One of the major subtypes of MT1. It has a wide range of biological characteristics, in addition to a detoxification of heavy metals and trace elements in the storage, transport and metabolism, but also has ability to enhance the body’s stress, antagonism of ionizing radiation, scavenging hydroxyl free radicals, regulation of cell proliferation and apoptosis function of the activity [12]. In recent years, MT and liver fibrosis has aroused widespread attention, that may be involved in the process of hepatic fibrosis, and hepatic stellate cells ( hepatic stellate cells, HSCs are the key objects of liver fibrosis [35]. There is now on the MT activity in the HSCs of the physiological and pathological role in the process of what, exactly, are poorly understood. In this study, recombinant eukaryotic expression plasmid was constructed MT1A pc
