Method of in vitro human neutrophil divided into saline control group (A group), to Cape amphotericin B (LMB) intervention group (B group), LPS stimulation group (C group) and LMB + LPS group (D group). After stimulation of each group were 120 min, with the Western blot nucleus, cytoplasm IB content of kit with NF Bp65 nuclear NF B activity detected by flow cytometry the percentage of neutrophil apoptosis. Results C Group IB content in cytoplasm and nucleus than in group A was significantly reduced, while NF B activity was significantly increased, reducing the proportion of neutrophil apoptosis; D group nucleus IB levels increased significantly compared with C group, NF B activity was significantly inhibited neutrophil apoptosis increased in proportion. Conclusion The accumulation of the nuclear IB can inhibit NF B activity, and to promote the inflammatory state of neutrophil apoptosis. After 120 min of stimulation, Western blot was applied to detect IBcontent in the nucleus and cytoplasm, NF B activity of the nucleus was detected by NF Bp65 kit, neutrophil apoptosis percentage was determined by flow cytometer.
