Methods: subcultured mesangial cells to take 4th generation in mesangial cells in experimental groups, using RT-PCR in each group were measured COX-2 mRNA and Smad2 mRNA expression, the effects of different concentrations of TGF-β at 4,12,24 h on the COX-2 mRNA and Smad2 mRNA expression, and its inhibitory effect of NS-398. Results: Different concentrations of TGF-β at different time, the mesangial cells and COX-2 mRNA expression of Smad2 mRNA increased in a concentration and time-dependent. Compared with the TGF-β by adding COX-2 inhibitor NS-398 after, COX-2 mRNA and Smad2 mRNA expression were reduced. Conclusion: TGF-β to stimulate mesangial cell expression of COX-2 signaling pathway is one of Smad signaling pathway. The COX-2 inhibitor NS-398 Dui inhibition of COX-2 expression in one of the mechanisms is through this signaling pathway.
