PAGE \* MERGEFORMAT 13 Penetration enhancers on in vitro artesunate transdermal patches of Of: Qiu Lin, Wang Naijie, Du Feng, Liang Bingwen, Ye Zuguang [Abstract] Objective To study the several penetration enhancers to artesunate in vitro percutaneous absorption, for the development of artesunate reference patches. Methods TK-6A transdermal diffusion device, the SD rat abdominal the skin barrier for the penetration measured penetration enhancers with different concentrations of artesunate by the amount of cumulative patches and percutaneous permeation rate. Results of penetration enhancers on the penetration effect of artesunate in the order: oleic acid ( OA)gt; N-methyl pyrrolidone (NMP)gt; propylene glycol (PG)gt; Azone (Azone)gt; adipate Isopropyl (IPA). NMP and permeability-promoting effect of OA concentration dependent, IPA and PG, penetration effect is not enhanced with the increase of concentration, high concentration of IPA prescription and PG but will inhibit the absorption of artesunate transdermal. Conclusion 10% OA to artesunate, the effect of the most significant penetration. [Keywords:] artesunate transdermal patches, percutaneous absorption, penetration agents, rats Abstract: Objective To investigate the effect of penetration enhancers on the percutaneous absorption of artesunate, and provide a reference for the development of artesunate transdermal delivery systems. Methods Penetration enhancers (in different concentrations) were incorporated into a 25% w / w artesunate matrix patch formulation. Modified Franz diffusion chambers and SD rat abdominal skin were used for in vitro skin permeation studies, the transdermal flux and permeation rate of artesunate in vitro were determined by HPLC. Result The enhancing effectiveness of the enhancers decreased in the order: OAgt; NMP gt; PGgt; Azonegt; IPA. Skin permeation was significantly enhanced by increasing the loading concentration of OA or NMP, larger IPA amount (8%) or PG amount (20%)
