Kurokawaetal.WorldJournalofSurgicalOncology2012,10:99 /content/10/1/99 WORLD JOURNAL OF SURGICAL ONCOLOGY RESEARCH OpenAccess EffectofL-argininesupplementonliver regenerationafterpartialhepatectomyinrats TsuyoshiKurokawa1,6*,JunAn ,KojiTsunekawa ,YoshiharuShimomura ,ShunsukeKazama ,NaohisaIshikawa3, 1,2 3,4 5 5 1 ToshiakiNonami andSatoruSugiyama4 Abstract Background:Nitricoxide(NO)hasbeenreportedtobeakeymediatorinhepatocyteproliferationduringliver regeneration.NOistheoxidativemetaboliteofL-arginine,andisproducedbyafamilyofenzymes,collective termednitricoxidesynthase(NOS).Thus,administrationofL-argininemightenhanceliverregenerationaftera hepatectomy.Anotheraminoacid,L-glutamine,whichplaysanimportantroleincatabolicstatesandisacrucial factorinvariouscellularandorganfunctions,iswidelyknowntoenhanceliverregenerationexperimentally.Thus, thepresentstudywasundertakentoevaluatetheeffectsofanL-argininesupplementonliverregeneration,andto comparedthiswithsupplementationwithL-glutamineandL-alanine(thelatterasanegativecontrol),usingarat partialhepatectomymodel. Methods:Beforeandaftera70%hepatectomy,ratsreceivedoneofthreeaminoacidsolutions(L-arginine, L-glutamine,orL-alanine).Theeffectsonliverregenerationoftheadministeredsolutionswereexaminedby assessmentofrestitutedlivermass,stainingforproliferatingcellnuclearantigen(PCNA),andtotalRNAandDNA content24and72hoursaftertheoperation. Results:At72hoursafterthehepatectomy,therestitutedlivermass,thePCNAlabelingindexandtheDNA quantitywereallsignificantlyhigherintheL-arginineandL-glutaminegroupsthaninthecontrol.Therewereno significantdifferencesinthoseparametersbetweentheL-arginineandL-glutaminegroups,norwereany significantdifferencesfoundbetweentheL-alaninegroupandthecontrol. Conclusion:OralsupplementsofL-arginineandL-glutamineenhancedliverregenerationafterhepatectomyin rats,suggestingthatanoralargininesupplementcanclinicallyimproverecoveryafteramajorliverresection.
