AnticancerActivityofAmaurodermarude ChunweiJiao1,2.,Yi-ZhenXie1.,XianglingYang2,HaoranLi2,3,Xiang-MinLi1,Hong-HuiPan1 ,Mian- HuaCai1,Hua-MeiZhong1,BurtonB.Yang2,3* 1Guangdong Institute of Microbiology, Guangzhou, China, 2Sunnybrook Research Institute, Sunnybrook Health Sciences Centre, Toronto, Canada, 3Department of LaboratoryMedicineandPathobiology,UniversityofToronto,Toronto,Canada Abstract More and more medicinal mushrooms have been widely used as a miraculous herb for health promotion, especially by cancerpatients.Herewereportscreeningthirteenmushroomsforanti-cancercellactivitiesinelevendifferentcelllines.Of theherbalproductstested,wefoundthattheextractofAmaurodermarudeexertedthehighestactivityinkillingmostof thesecancercelllines.AmaurodermarudeisafungusbelongingtotheGanodermataceaefamily.TheAmaurodermagenus containsapproximately30specieswidespreadthroughoutthetropicalareas.SincethebiologicalfunctionofAmauroderma rudeisunknown,weexamineditsanti-cancereffectonbreastcarcinomacelllines.Wecomparedtheanti-canceractivityof Amauroderma rude and Ganoderma lucidum, the most well-known medicinal mushrooms with anti-cancer activity and found that Amauroderma rude had significantly higher activity in killing cancer cells than Ganoderma lucidum. We then examinedtheeffectofAmaurodermarudeonbreastcancercellsandfoundthatatlowconcentrations,Amaurodermarude couldinhibitcancercellsurvivalandinduceapoptosis.Treatedcancercellsalsoformedfewerandsmallercoloniesthanthe untreatedcells.WhennudemicebearingtumorswereinjectedwithAmaurodermarudeextract,thetumorsgrewataslower ratethanthecontrol.Examinationofthesetumorsrevealedextensivecelldeath,decreasedproliferationrateasstainedby Ki67,andincreasedapoptosisasstainedbyTUNEL.Suppressionofc-mycexpressionappearedtobeassociatedwiththese effects.Takentogether,Amaurodermaruderepresentedapowerfulmedicinalmushroomwithanti-canceractivities.
