AntidiabeticandAntioxidantEffectsandPhytochemicals ofMulberryFruit(MorusalbaL.)PolyphenolEnhanced Extract YihaiWang,LiminXiang,ChunhuaWang,ChaoTang,XiangjiuHe* SchoolofPharmaceuticalSciences,WuhanUniversity,Wuhan,China Abstract Theantidiabeticandantioxidantactivitiesoftheethylacetate-solubleextract(MFE)ofmulberryfruit(MorusalbaL.)were investigated.Invitro,MFEshowedpotenta-glucosidaseinhibitoryactivityandradical-scavengingactivitiesagainstDPPH andsuperoxideanionradicals.Invivo,MFEcouldsignificantlydecreasefastingbloodglucose(FBG)andglycosylatedserum protein (GSP), and increase antioxidant enzymatic activities (SOD, CAT, GSH-Px) in streptozotocin (STZ)-induced diabetic mice.Bioactivity-guidedfractionationoftheMFEledtotheisolationof25phenoliccompounds,andtheirstructureswere identifiedonthebasisofMSandNMRdata.Allthe25compoundswereisolatedfrommulberryfruitforthefirsttime.Also, the a-glucosidase inhibitory activity and antioxidant activity of the phenolics were evaluated. Potent a-glucosidase inhibitory and radical-scavenging activities of these phenolics suggested that they may be partially responsible for the antidiabeticandantioxidantactivitiesofmulberryfruit.
